En 1947, à Chypre. Des milliers de réfugiés juifs en route pour la Terre sainte sont interceptés par les Anglais et parqués dans des camps. Craignant des heurts avec la population arabe, les autorités britanniques refusent de les laisser rejoindre la Palestine. Ari Ben Canaan, un agent de l'organisation sioniste clandestine Haganah, réussit à se faire passer pour un officier anglais. Il embarque les derniers arrivants sur un vieux bateau grec rebaptisé "Exodus". Défiant les autorités britanniques, Ari menace de faire sauter le navire si celles-ci ne lèvent pas le blocus. L'infirmière Kitty Fremont, témoin de la manière dont sont traités les juifs, sympathise avec la cause sioniste...
